A long term study of bonamiosis in Quiberon bay, France ArchiMer

Bonamiosis was first reported in association with mass mortality of flat oysters, Ostrea edulis, in June 1979 in L'Ile Tudy, Brittany. The disease rapidly spread to all of the oyster farming areas in France but also in other European countries. The French flat oyster production, which already suffered from another protozoan disease, marteiliosis, decreased from 5500 mt in 1979 to less than 2000 mt after 1980. In 2001, about 350 French farms sell 1650 mt of flat oysters. This production mainly relies on natural spat collection which specially occurs in Quiberon bay. Arbitrarily primed PCR to type Vibrio spp. Pathogenic for shrimp ArchiMer

A molecular typing study on Vibrio strains implicated in shrimp disease outbreaks in New Caledonia and Japan was conducted by using AP-PCR (arbitrarily primed PCR). It allowed rapid identification of isolates at the genospecies level and studies of infraspecific population structures of epidemiological interest. Clusters identified within the species Vibrio penaeicida were related to their area of origin, allowing discrimination between Japanese and New Caledonian isolates, as well as between those from two different bays in New Caledonia separated by only 50 km. Le réseau REPAMO (Réseau de Pathologie des Mollusques) est un réseau de santé animale, qui s'occupe des coquillages marins du littoral français métropolitain. Les mollusques marins des côtes françaises représentent une ressource par la pêche, mais sont surtout une part importante de l'aquaculture. Les pertes liées aux maladies dans ce domaine d'activité sont aujourd'hui considérées comme l'un des principaux facteurs limitants de son développement au niveau mondial. La traduction en droit français des Directives Européennes 91167/CE et 95/70/CE confère au Réseau de Pathologie des Mollusques de l'IFREMER (REPAMO) la charge de contrôler la santé des invertébrés marins d' intérêt commercial sur le territoire national. A parasite of the dredge oyster Ostrea chilensis Philippi, previously known as Bonamia sp., was recently described and named Bonamia exitiosus, based on ultrastructural and molecular evidence. The rules governing the names of organisms require correspondence of gender, and the name must be changed from B. exitiosus to B. exitiosa.

Bonamia sp. is a pathogenic parasite that occurs in the haemocytes of dredge oysters Ostrea chilensis Philippi in New Zealand, Ultrastructurally it resembles other haplosporidians in the possession of haplosporosomes, haplosporogenesis, persistence of mitotic microtubules during interphase and of the nuclear envelope during mitosis, and occurrence of a diplokaryotic or multi-nucleate plasmodial stage. Another stage containing a large vacuole derived from enlargement of 1 or more mitochondria has not previously been described from other haplosporidians. It most closely resembles B, ostreae Pichot et al., 1979, which parasitises and is pathogenic in haemocytes of European flat oysters, O. edulis. Nowadays, cupped oyster, Crassostrea gigas, production is the main shellfish production in France. Spats cultivated on most farms are usually collected in the wild, but hatchery spats are becoming more numerous, and represent around 10% of today's cultivated spats. During the summer period, when the temperatures are higher, high mortality rates have been regularly observed in cupped in oyster C. gigas (60 to 100%) spats since 1991, both in hatcheries-nurseries and in their natural habitat. Over the last five years, these episodes have become a big concern for all the professionals.

Sample-size calculation in the context of the surveys aimed at substantiating freedom from infection have been commonly undertaken an terrestrial animals over recent years, but not on aquatic animals. A recent model developed by Audigé and Beckett in 1999 can be used to plan and assess animal health surveys. The aim of this study was to adapt that model for marine aquaculture, in particular to help in planning surveys aimed at substantiating freedom form two exotic diseases, mikrocytosis and perkinsosis, in the French population of Crassostrea gigas. We isolated complementary DNA (Cg-TIMP) from Crassostrea gigas that codes for a protein highly homologous to vertebrate tissue inhibitors of metalloproteinases (TIMPs). In vertebrates, TIMPs are multifunctional proteins that play a key role in extracellular matrix metabolism. They are involved in physiological processes (e.g. embryonic development, damage repair, angiogenesis) and pathological processes (e.g. inflammatory reactions, cancer).

Comparative analysis of Vibrio splendidus-related strains isolated during Crassostrea gigas mortality events ArchiMer

French mollusc production is based mainly on the Pacific cupped oyster, Crassostrea gigas. Since 1991, annual mass mortality of juveniles has been reported during summer months. These recurring episodes concern professionals who fear that like Portugese oyster, C. angulata, C gigas could in turn disappear following one of these epizooties. Previously, bacteriological analysis of moribund oyster juveniles yielded an isolate of a Vibrio splendidus biovar H strain, named TNEMF6. Bonamiosis refers to diseases of oysters caused by Bonamia ostreae in the Northern Hemisphere and Bonamia sp. in the Southern Hemisphere. Similarly, Mikrocytosis usually refers to diseases of oysters caused by Mikrocytos mackini on the west coast of Canada and Mikrocytos roughleyi on the east coast of Australia.

Marteilia spp. are paramyxean parasites that affect several bivalve species of economic interest, such as Ostrea edulis and Mytilus galloprovincialis. Certain aspects of Marteilia spp., such as their life cycle and host affinity and infection dynamics, still remain unknown. Development of a TaqMan PCR assay for the detection of Bonamia species ArchiMer

The development of molecular diagnostic assays with increased sensitivity compared with conventional histological techniques is highly desirable for effective management of bonamiosis in cultured oyster stocks and wild populations. A real-time TaqMan PCR assay was developed for the specific detection of Bonamia species in infected oyster tissues. The TaqMan assay was shown to be significantly more sensitive than histopathology. Although a real-time TaqMan PCR assay is comparable with conventional PCR in terms of sensitivity, it offers the advantages that it is a rapid test and has a very low risk of sample cross-contamination. Diseases in mollusc hatcheries and their paraedox in health management ArchiMer

Global molluscan aquaculture production is continuously increasing, dominated by five species, among which the Pacific cupped oyster, Crassostrea gigas, predominates. Hatchery production is accompanying this increase to assist consistent availability of juveniles for restocking, fishery enhancement, genetic improvements as well as for species diversification. Hatchery development contributes significantly to the demand for international transfers of live molluscs; a consequence of which pathogen transfer via transfer of live molluscs is currently recognised as a major cause of epizootic disease outbreaks.
